Telomeres in Arabidopsis thaliana plants treated by hypomethylation drugs

Abstract:

Epigenetics deals with the study of modifications of nucleic acids and histones and with the involvement of these modifications in regulation processes. Methylation of DNA at the carbon 5 of the cytosine residue is important and frequently studied epigenetic modification. Telomeres, nucleoprotein structures located at the ends of eukaryotic chromosomes, are also subjects of epigenetics modifications. Telomeres of the model plant Arabidopsis thaliana consist of short repetitive sequences TTTAGGG, and cytosines in the complementary AAATCCC strand can be methylated. In the theoretical part of this bachelor thesis, data on epigenetic modifications, with an emphasis on DNA methylation and hypomethylation drugs used for functional analyses are summarised. In the experimental part, telomeres in three ecotypes of A. thaliana, with different telomere lengths germinated in the presence of hypomethylation drug zebularine were analysed. Although data on respective change of telomere lengths in hypomethylated plants were not obtained due to the experimental imprecision, we have got technical information important for following analyses. Finally, results of analogous experiments using model plants A. thaliana and Nicotiana tabacum are discussed.

Abstract:

Epigenetika se zabývá studiem modifikací nukleových kyselin a histonů a úlohou těchto modifikací v regulačních procesech. Mezi nejprostudovanější epigenetické modifikace patří metylace cytozinu na uhlíku v poloze 5. Epigenetickým modifikacím podléhají i koncové části eukaryotických chromozomů, telomery, skládající se z krátkých repetitivních sekvencí DNA a specifických a nespecifických proteinů včetně histonů. Telomery rostliny Arabidopsis thaliana jsou tvořeny opakující se sekvencí TTTAGGG, přičemž cytosiny v komplementárním vlákně CCCTAAA mohou být metylovány. V teoretické části bakalářské práce jsou shrnuty poznatky o epigenetických modifikacích, zejména metylaci DNA a hypometylačních činidlech, která se využívají při studiu její funkce. Dále se teoretická část věnuje telomerám a zapojení epigenetických mechanismů do jejich regulace. V praktické části se práce zabývá analýzou telomer ve třech ekotypech A. thaliana s různou délkou telomer, které klíčily v přítomnosti hypometylační látky zebularinu. Ačkoliv se kvůli chybě v provedení experimentu nepovedlo získat data o případné změně délky telomer v hypometylovaných rostlinách, získali jsme významné technické informace pro další analýzy. Diskuse se věnuje podobným experimentům provedeným na modelových rostlinách A. thaliana a Nicotiana tabacum.
