Methods for immobilization of proteins on the surface of microtitration plates

Abstract:

Cílem této bakalářské práce bylo porovnat klasickou pasivní adsorbci s kovalentní imobilizací proteinů na povrch různých mikrotitračních destiček a stripů. Dále byla porovnána vazba na povrch destičky potažený streptavidinem pro vazbu biotinylovaných molekul. Jako protilátka byla použita monoklonální protilátka AL01, albumin lidského séra a anti albumin. Nejprve bylo nutné protilátku naimobilizovat na povrch destiček. Protilátka byla zředěna karbonátovým pufrem na koncentrace 5g/ml a 20g/ml. Po inkubaci byla přidána enzymem značená sekundární protilátka IgG-HRP a HSA-HRP, která se navázala na protilátku. Množství navázaného konjugátu je pro tento test nejdůležitější. Peroxidáza byla kvantifikována přidáním 3,5,3‘,5‘-tetramethylbenzidinu jako substrátu s peroxidem vodíku. Enzymová reakce byla zastavena 2,5 M HCl a absorbance byla měřena při 450 nm. Absorbance je přímo úměrná množství navázaného konjugátu. Porovnáním všech naměřených absorbancí bylo zjištěno, že největší enzymovou aktivitu, tedy největší množství navázaného konjugátu, vykazuje mikrotitrační destička GAMA, pasivní adsorbce. Naopak nejnižší hodnoty absorbance byly naměřeny na destičce Nunc polysorb/maxisorb, pasivní adsorbce. Mírně vyšší hodnoty vykazovaly destičky Nunc Immobilizer Amino, kovaletní imobilizace, a Nunc Immobilizer Streptavidin, afinitní interakce. Nižší absorbance těchto destiček mohly být způsobeny velmi silnou enzymovou reakcí; při zastavení reakce došlo ke sražení barvy. To mohlo ovlivnit výsledky měření.

Abstract:

A target of this bachelor thesis was comparison of a classical passive coupling with covalent bonding of proteins on a surface of different microtiter plates and strips. The streptavidin coated plates was tested, this surface couples biotinylated molecules. The model interaction was represented by human serum albumin and anti albumin monoclonal antibody AL01. At first it was necessary to imobilize the antibody AL01 to the surface of microwells. Antibody was diluted in coating buffer at two different concentrations, 5g/ml and 20g/ml. Subsequently, after incubation and washing, enzym linked secondary antibody IgG-HRP or HSA-HRP conjugate was added, which was coupled to the primary antibody. Quantity of binding conjugate was the most important for this immunoassay. The surface-bound peroxidase was quantified using 3,5,3‘,5‘-tetramethylbenzidine as substrate with hydrogen peroxide. The enzym reaction was stopped by 2,5 M HCl and absorbance was measured at 450nm. Absorbance was directly proportional to quantity of the bound conjugate. By comparison of all measured absorbances of all microtiter plates and strips, it was detected that the most coupled enzyme activity and thus the best immobilization effectiveness was exhibited by the microtiter plate GAMA with passive coupling. On the contrary, the lowest value of absorbace were measured on the microtiter plate Nunc polysorb/maxisorb, based on the passive coupling, too. Moderately high responses were obtained with covalent coupling of Nunc Immobilizer Amino and affinity coupling of Nunc Immobilizer Streptavidin. A lower absorbances of these microtiter plates could be caused by very strong enzyme reaction; at stopping, this reaction resulted in partial precipitation of color. Thus this complication could have influenced results of measurements.
