Factors affecting numbers of mitochondria in rodents

Abstract:

Mitochondrie jsou buněčné organely zodpovědné za syntézu ATP. Změny v podmínkách prostředí mohou u mitochondrií vyvolat adaptivní změny v jejich struktuře nebo počtu. Takovéto změny mohou vyústit například ve zvýšenou produkci ATP, což se zdá být výhodou při adaptacích na vysokou nadmořskou výšku. Rešeršní část bakalářské práce se věnuje stručnému popisu mitochondrií a jejich energetického metabolismu. Dále popisu faktorů prostředí, které nejčastěji ovlivňují množství, strukturu a vlastnosti mitochondrií, a evolučních procesů pozitivní selekce a mitochondriální introgrese, které k těmto změnám mohou vést. Součástí literární rešerše je také krátké shrnutí možných metod kvantifikace mitochondrií a podrobněji popsaná metoda využitá v praktické části této práce. Praktická část se věnuje kvantifikaci mitochondrií u hlodavců z rodů Stenocephalemys a Lophuromys z etiopských pohoří Guassa a Choqa za využití kvantitativní PCR a následné statistické analýzy. Překvapivě se nepotvrdil vliv nadmořské výšky na počet mitochondrií, což může být způsobeno možnou krátkodobou změnou počtu mitochondrií nikoli změnou trvalou, která přetrvává po další generace. Množství mitochondrií se signifikantně lišilo mezi jednotlivými typy tkání.

Abstract:

Mitochondria are cellular organels responsible for ATP production. Changes in environmental conditions can induce adaptive changes in mitochondrial structure or their numbers. Such changes can result, for example, in increased ATP production, which appears to be an advantage in adaptations to high altitude. The review part of the bachelor thesis is devoted to a brief description of mitochondria and their metabolism. Futhermore, I describe the environmental factors most commonly affecting the number, structure and function of mitochondria, and the evolutionary processes of positive selection and mitochondrial introgression that might lead to these changes. The practical part deals with the quantification of mitochondria in rodents from the genera Stenocephalemys and Lophuromys from the Guassa and Choqa mountain ranges of Ethiopian Highlands, using quantitative PCR and subsequent statistical analysis. Suprisingly, the effect of elevation on mitochondrial quantity was not confirmed, which may be due to possible short-term change in mitochondrial number rather than a permanent change that persists over generations. The number of mitochondria differed significantly between tissue types.
