Synthesis and characterization of anticholineric carbamates containing benzothiazole block

Abstract:

Tématem diplomové práce je syntéza a následná charakterizace nepopsaných anticholinergních karbamátů obsahujících benzthiazolový blok. V teoretické části byla provedena rešerše, jejímž cílem bylo nalézt vhodnou syntézu karbamátů s výše zmíněným benzthiazolovým blokem. Literární rešerše byla také zaměřena na studie stanovující inhibiční aktivitu anticholinergních sloučenin s benzthiazolovým blokem. Poznatky byly dále využity při syntéze vybraných sloučenin. Na základě vypracované literární rešerše bylo v experimentální části připraveno 18 nových karbamátů. Připravené karbamáty byly charakterizovány NMR spektroskopií, bodem tání, HRMS MALDI a elementární analýzou. Na závěr byla u těchto karbamátů stanovena inhibiční aktivita proti acetylcholinesteráze a butyrylcholinesteráze. Také byla stanovena lipofilita připravených karbamátů. Zjištěním míry lipofility připravených karbamátů lze dokázat schopnost sloučeniny přecházet přes lipofilní hematoencefalickou bariéru.

Abstract:

The topic of this diploma thesis is the synthesis and subsequent characterization of non-described anticholinergic carbamates containing benzthiazole block. In the theoretical part, a search was carried out to find a suitable carbamate synthesis with the aforementioned benzthiazole block. The literature search was also focused on studies determining the inhibitory activity of anticholinergic compounds with the benzthiazole block. The findings were further used in the synthesis of selected compounds. Based on the literature search, 18 new carbamates were prepared in the experimental part. The prepared compounds were characterized by NMR spectroscopy, melting point, HRMS MALDI and elemental analysis. Finally, acetylcholinesterase and butyrylcholinesterase inhibitory activity was determined for these carbamates. Lipophilicity of prepared carbamates was also determined. By determining the lipophilicity of the prepared carbamates, the ability of the compound to cross the lipophilic blood-brain barrier can be demonstrated.
