Diversity and distribution of parasitic digeneans in European Pelophylax water frogs

Abstract:

Although the species distribution of water frogs in the Western Palaearctic is relatively well researched, very little is known about the species richness of the parasite fauna of individual species. The proposed thesis expands knowledge about the species diversity of trematodes parasitizing water frogs P. esculentus and P. ridibundus from southern Slovakia and provides the first information about the parasite fauna of endemic water frogs P. epeiroticus and P. kurtmuelleri from the Balkan Peninsula. A total of 91 water frogs were examined, of which 28 were collected from Balkan localities and 63 from Slovakia. Altogether, more than 2,200 trematodes belonging to 21 species were found in host frogs. Seventeen species of trematodes were found in water frogs from Slovakia, while 12 species parasitized endemic water frogs from Balkan localities. 38% of all species parasitized in water frogs from both geographical areas. A combination of morphological analysis and molecular data was used to identify individual species, which revealed six potentially new species of trematodes. In order to quantify parasitic infection, basic epidemiological parameters were determined for individual species of trematodes, and diversity indices were calculated to assess alpha diversity. The highest mean prevalence was reported for Opisthioglyphe ranae found in the intestine of frogs. A significant output of the thesis is a phylogenetic analysis using partial 28S rDNA, which divided the found species of trematodes into five well-supported groups. In selected taxonomic groups, phylogenetic analysis was performed using the mitochondrial COI gene to reveal intraspecific variability. Molecular data provide a new perspective on the taxonomy of frog trematodes, and it is clear the diversity of this group in Europe is still not sufficiently studied.

Abstract:

I přesto, že druhová distribuce vodních skokanů v západním palearkte je relativně dobře prozkoumána, o druhové bohatosti parazitofauny jednotlivých druhů se ví jen velmi málo. Diplomová práce rozšiřuje poznatky o druhové rozmanitosti motolic parazitujících ve vodních skokanech P. esculentus a P. ridibundus z jižního Slovenska a zároveň přináší první informace o parazitofaune endemických vodních skokanů P. epeiroticus a P. kurtmuelleri z Balkánského poloostrova. Celkem bylo vyšetřeno 91 vodních skokanů, z toho 28 z balkánských lokalit a 63 ze Slovenska. Dohromady bylo v jedincích nalezeno přes 2200 motolic patřících do 21 druhů. Ve skokanech ze Slovenska bylo nalezeno 17 druhů motolic, zatímco v endemických skokanech z balkánských lokalit parazitovalo 12 druhů motolic. Z celkového počtu, 38% všech druhů motolic parazitovaly v skokan z obou geografických oblastí. K identifikaci jednotlivých druhů byla použita kombinace morfologické analýzy a molekulárních dat, což umožnilo odhalit 6 potenciálně nových druhů motolic. Za účelem kvantifikace parazitární infekce byly pro jednotlivé druhy motolic stanovené hodnoty základních epidemiologických parametrů a k hodnocení alfa diverzity byly vypočteny indexy diverzity. Nejvyšší průměrnou prevalenci dosáhl druh Opisthioglyphe ranae z tenkého střeva žab. Důležitým výstupem diplomové práce je také fylogenetická rekonstrukce za použití parciálního genu 28S rDNA, která rozdělila nalezené druhy motolic do 5 dobře podpořených skupin. U vybraných taxonomických skupin proběhla fylogenetická analýza na základě mitochondriálního genu COI za účelem odhalení vnitrodruhové variability. Molekulární data přinášejí nový pohled na taxonomii motolic žab a je zřejmé, že diverzita této skupiny v Evropě není stále dostatečně prozkoumaná.
