Mass spectrometry analysis of extracellular vesicles contained in ovarian cancer ascites

Abstract:

Rakovina vaječníků (OvCa) je heterogenní onemocnění vyznačující se špatnou prognózou a vysokou mortalitou způsobenou diagnostikou onemocnění v pokročilém stádiu. Extracelulární vesikuly (EVs) jsou membránové váčky, které jsou přítomny v tělních tekutinách, kde hrají významnou roli v mezibuněčné komunikaci, a zároveň díky tomu mohou být využity pro diagnostiku/monitorování různých onemocnění. Tato diplomová práce se zaměřuje na identifikaci specifického proteinu na povrchu malých extracelulárních vesikulů (sEVs), který by mohl být použitý jako potenciální biomarker pro včasnou diagnostiku OvCa. Metody založené na chromatografii využívající rozdílné velikosti částic a ultracentrifugaci byly použity pro izolaci sEVs z biologických tekutin a kondiciovaného média. Následná analýza pomocí hmotnostní spektrometrie identifikovala rozdíly mezi proteomickým profilem sEVs izolovaných z ascitu pacientek s OvCa a z tekutin zdravých jedinců. Jeden specifický protein byl identifikován v ascitu a plazmě analyzované skupiny onkologických pacientek, čímž byl položen základ pro další zkoumání tohoto potenciálního biomarkeru pro časnou detekci OvCa.

Abstract:

Ovarian cancer (OvCa) is a heterogeneous disease with a very poor prognosis and high mortality rate related to its diagnosis at advanced stages. Extracellular vesicles (EVs) are circulating microparticles that regulate communication between cells and can be also employed as disease biomarkers. This diploma thesis is focused on the identification of a specific protein on small extracellular vesicles (sEVs), which could be used as a potential biomarker for early detection of OvCa. Size exclusion chromatography and ultracentrifugation methods were evaluated for isolation of sEVs from several biological fluids and conditioned media. Consecutive mass spectrometry analysis allowed the identification of cancer related differences between proteomic profiles of sEVs isolated from ascites of OvCa patients and fluids of healthy individuals. One specific protein was identified in both the ascites and plasma of the analysed group of oncological patients. This finding laid foundations for further investigation of the identified protein as a potential biomarker for early detection of OvCa.
